Melatonin Enhances Object Recognition Memory through Melatonin MT1 and MT2 Receptor-Mediated and Non-Receptor-Mediated Mechanisms in Male Mice

چکیده
Melatonin (MEL) has been reported to have acute enhancing effects on some aspects of cognition. Recently, we revealed that N1-acetyl-5-methoxyquinuramine (AMK), a brain metabolite MEL, is much more potent than MEL in converting short-term memory (STM) long-term (LTM) with single administration immediately after the acquisition trial novel object recognition (NOR) task. These data suggest memory-enhancing may be mediated by mechanisms independent activation MT1 and MT2 receptors. In present study, examined contribution receptor-mediated non-receptor-mediated using NOR Mice were administered either AMK, or highly selective MT1/MT2 receptor agonist ramelteon (RAM) varying doses these drugs both STM LTM performance compared. We found AMK RAM facilitating promoting formation. also pretreatment luzindole, antagonist, markedly suppressed only RAM. results acutely enhances through mechanisms.

Melatonin transmits photoperiodic signals through the MT1 melatonin receptor.
Melatonin transmits photoperiodic signals that regulate reproduction. Two melatonin receptors (MT1 and MT2) have been cloned in mammals and additional melatonin binding sites suggested, but the receptor that mediates the effects of melatonin on the photoperiodic gonadal response has not yet been identified. We therefore investigated in mice whether and how targeted disruption of MT1, MT2, or bo...
متن کاملThe melatonin MT1 receptor axis modulates mutant Huntingtin-mediated toxicity.
Melatonin mediates neuroprotection in several experimental models of neurodegeneration. It is not yet known, however, whether melatonin provides neuroprotection in genetic models of Huntington's disease (HD). We report that melatonin delays disease onset and mortality in a transgenic mouse model of HD. Moreover, mutant huntingtin (htt)-mediated toxicity in cells, mice, and humans is associated ...
متن کاملSelective MT2 melatonin receptor antagonists block melatonin-mediated phase advances of circadian rhythms.
This study demonstrates the involvement of the MT2 (Mel1b) melatonin receptor in mediating phase advances of circadian activity rhythms by melatonin. In situ hybridization histochemistry with digoxigenin-labeled oligonucleotide probes revealed for the first time the expression of mt1 and MT2 melatonin receptor mRNA within the suprachiasmatic nucleus of the C3H/HeN mouse. Melatonin (0.9 to 30 mi...

Recent studies have demonstrated that the human placenta is a novel source of adult stem cells. We have provided laboratory evidence that transplantation of these human placenta-derived cells in vitro and in vivo stroke models promotes functional recovery. However, the mechanisms underlying these observed therapeutic benefits of human placenta-derived cells unfortunately remain poorly understoo...
متن کاملCircadian Pattern of Melatonin MT1 and MT2 Receptor Localization in the Rat Suprachiasmatic Nucleus.
The suprachiasmatic nucleus (SCN) is the master circadian pacemaker. The pineal hormone melatonin is involved in the regulation of circadian phase. As a part of the circadian system, its synthesis and secretion is under SCN control. On the other hand, melatonin feeds back on the SCN to regulate its function. Melatonin has two specific windows of time at which it regulates SCN function, namely d...
